An ultrastructural comparison of blackspot bruise and shatter bruise in potato (Solanum tuberosum) tubers.

Potatoes cv. Pentland Dell tubers were impacted with a falling bolt at harvesting at ADAS Arthur Rickwood, Cambridgeshire, UK or at 12-weekly intervals thereafter through storage. Impacted and control tubers were incubated at 22 deg C for 48 h before sampling for ultrastructural analysis. Tubers impacted at harvest developed microscopic shatter compared with stored tubers which formed blackspot bruise. Microscopic shatter was characterized as a disrupted tonoplast showing vesicularization, increased numbers of mitochondria and nuclear invaginations. In severe cases, tissue damage resulted in cavities forming within the cells. Conversely, blackspot formation typically appeared as melanin formation surrounding the starch grains and adjacent to the cell wall.
