Collection and analysis of headspace volatiles for disease detection in stored potatoes.

 

Volatile production by stored potatoes infected with a specific bacteria (Erwinia carotovora) was analysed by means of gas chromatography. The potatoes were inoculated with the bacterial culture and stored at 12 deg C in sealed containers. A flow of purified air was passed through the containers and subsequently through glass tubes (traps) filled with a solid adsorbent (Chromosorb 105). This adsorbent was selected due to its high ability to trap organic compounds. The duration of the experiment was 15 d, during which the traps were replaced at 24-h intervals. The trapped volatile compounds were thermally desorbed and analysed by gas chromatography. The progression of volatile compounds by healthy and diseased tubers was compared. Results show that the total amount of volatile production by infected potatoes was significantly higher than that of healthy tubers and at least 1 volatile compound was specific to the disease.
