Fluorescence polarization as a means for determination of fumonisins in maize.

A rapid, portable fluorescence polarisation based assay for fumonisins in maize is described. The assay is based on the competition of unlabelled fumonisin from a sample, with a fluorescently tagged fumonisin (FB1 – FL) for a frumonisin – specific monoclonal antibodyin solution. The fluorescence polarisation of the tagged fumonisin is increased upon binding with the antibody. In the presence of free toxin, less of the FB1 – FL si bound and the polarisation signal is decreased. The assays were very simple to perform, requiring only mixing of and aqueous extract of maize with the tagged fumonisin and antibody, and required less than 2 minutes per sample, excluding extraction time. The limit of detection for FB1 was 0.5 mg/kg in spiked maize. Forty – eight samples offield – contaminated maize were tested by the fluorescence po;larisation and an extablished HPLC method, with a good corredlation between the two.
