Determination of the mycotoxin moniliformin in cultures of Fusarium subglutinans and in naturally contaminated mize by high-performance liquid chromatography-at-mospheric pressure chemical ionization mass spectrometry.

An LC-Ms method employing triethylamine as ion-pairing reagent for the determination of MON in culture material and naturally contaminated maize samples is described.  MS detection of MON was accomplished following atmospheric pressure chemical ionisation to yield the  deprotonated molecular ion [M-H] at m/z 97.  The MON response was linear over the injected range 10 to 700 ng and a detectionlimit of 10 ng was achieved.  Five Souht African strains of Fusarium subglutinans were grown on maize kernels and MON extracted with an acetonitrile-water (95:5) mixture.  Following sample cleanup with reversed phase SPE cartridges, the extracts were subjected to LC-MS analysis.  Triethylamine was used as anion-pari reagent and found to improve the retention characteristics of MON without anydetrimental effects to the instrument.  Application of this method to naturally contaminated maize samples from Transkei showed  that it was capable of measuring Mon levels down to 10 ug/kg in mouldy maize cobs.
